How to renew a stem cell

چکیده

ike a picky tourist looking for a hotel room, the malariacausing protozoan Plasmodium passes through several cells before settling on one to infect. Margarida Carrolo, Maria Mota (Instituto Gulbenkian de Ciência, Oeiras, Portugal), and colleagues now report that the initial invasions cause the damaged cells to make a factor that primes other cells for infection. The factor turned up in culture medium from infected or wounded cells. When added to uninfected cells, this conditioned medium doubled the level of infection. The factor was identified as hepatocyte growth factor (HGF), which is made by hepatocytes in response to wounding. Those cells that were permeable to dextran (because of Plasmodium induced wounding) were the same ones that made HGF. Mota earlier showed that the passage through cells primes Plasmodium for infection. Once it is primed, Plasmodium induces a folding of the plasma membrane that becomes a parasitophorous vacuole. The Portugese group noted HGFinduced changes in the actin cytoskeleton in hepatocytes, and they speculate that these changes may be necessary for early parasite development. The HGF receptor Met is not a receptor for Plasmodium , but when the group inhibited Met’s kinase activity this did prevent Plasmodium infection, even when the inhibition occurred after invasion. Any such treatment “needs to be prophylactic, because people don’t know they are infected at this stage,” says Mota. She is contacting companies that are developing anti-cancer drugs targeting Met, and also hopes to find other targets downstream of Met activation.

Embryonic stem cells are pluripotent stem cells which have the ability to indefinitely self-renew and differentiate into all differentiated cells of the body. Regarding their two main properties (unlimited self-renewal and multi-lineage differentiation), these cells have various biomedical applications in basic research and cell based therapy. Because the transplantation of differentiated cells...

Maintenance of horse embryonic stem cells in different conditions

Embryonic stem cells (ESCs) are originally derived from the ICM of blastocysts and are characterized by their ability to self-renew and their pluripotencies. Only a few reports have been published on ESC isolations and line establishment in animals, even fewer in horses. However, it is still important to isolate equine ESCs for animal biotechnology and therapeutic applications. In the present s...

Stem Cell Therapy in Pediatric Neurological Disorders

Pediatric neurological disorders including muscular dystrophy, cerebral palsy, and spinal cord injury are defined as a heterogenous group of diseases, of which some are known to be genetic. The two significant features represented for stem cells, leading to distinguish them from other cell types are addressed as below: they can renew themselves besides the ability to differentiate into cells wi...

The Cancer Stem Cell Hypothesis in Oral Squamous Cell Carcinoma: A New Target for the Treatment

Within a single tumor clone, cells have significantly different abilities to proliferate and form new tumors. This has led to the hypothesis that most cells in a cancer have a limited ability to divide and only a small subset of distinct cells, the cancer stem cells (CSCs), has the capacity to self-renew and form new tumors . It has been proposed that the development of tumors is based exclusiv...

The Effects of Dental Pulp Stem Cell Conditioned Media on the Proliferation of Peripheral Blood Mononuclear Cells

Background: Dental Pulp Stem Cells (DPSCs) are multipotent mesenchymal stem cells. DPSCs can renew themselves and differentiate into various cell types such as adipocytes, osteocytes, neurons, etc. DPSCs possess immunomodulatory properties and can inhibit peripheral blood mononuclear cell (PBMC) proliferation. Recent studies showed that conditioned-medium mesenchymal stem cells also had immunos...

Brat is a Miranda cargo protein that promotes neuronal differentiation and inhibits neuroblast self-renewal.

An important question in stem cell biology is how a cell decides to self-renew or differentiate. Drosophila neuroblasts divide asymmetrically to self-renew and generate differentiating progeny called GMCs. Here, we report that the Brain tumor (Brat) translation repressor is partitioned into GMCs via direct interaction with the Miranda scaffolding protein. In brat mutants, another Miranda cargo ...
